As a sixteen-year-old high school student, McGuire finished fourth in the 100-yard dash at the 1960 AAU national championships. She entered Tennessee State University in the fall of 1961.
At the 1964 Olympics, McGuire won a gold medal in the 200-meter dash, running a 23.0 to break Wilma Rudolph's Olympic record. She finished second to college teammate Wyomia Tyus in the 100-meter and won a second silver medal as a member of the 4 by 100-meter relay team.
McGuire won the AAU national 100-meter in 1963 and was the 200-meter champion in 1964 and 1965. She also won a gold medal in the 100-meter at the 1963 Pan-American Games.
After graduating from college in 1966, McGuire retired from competition and became a teacher in Atlanta. She later worked with underprivileged children in Detroit.
